摘要
目的 研究人脑胶质瘤缺氧诱导因子 1α(HIF 1α)、CD34 表达与预后的关系。方法 采用免疫组织化学方法检测63例脑胶质瘤HIF 1α、CD34 的表达状况 ,并将结果与临床预后随访结果进行综合分析。结果 ①HIF 1α总阳性表达率为71 4% ,其中强表达 16例 ( 2 5 4% ) ,中度表达 14例 ( 2 2 2 % ) ,弱表达 15例 ( 2 5 4% ) ,不表达 18例 ( 2 8 6% ) ,有显著性差异(P <0 0 1)。HIF 1α表达水平与胶质瘤恶性程度相关 (P <0 0 1)。HIF 1α主要表达于肿瘤边缘或与正常组织交界处的细胞及坏死的肿瘤组织。胶质瘤Ⅰ级、Ⅱ级、Ⅲ~Ⅳ级微血管密度 (MVD)分别为 ( 3 5± 14 )、 ( 69± 3 3 )、 ( 119± 49) ,有显著差异(P <0 0 1)。②CD34 表达与HIF 1α表达水平相关 (P <0 0 1)。③HIF 1α表达与生存时间负相关 (P <0 0 1)。结论 CD34与HIF 1α表达有一定相关性 ,HIF 1α与脑胶质瘤新生血管生成有关。HIF 1α、CD34
Objective: To study the expressions of HIF-1α and CD 34 in gliomas and their correlations with prognosis. Methods: The expressions of HIF-1α and CD 34 were investigated by immunohistochemistry in 63 specimens of gliomas. Then the influence of HIF-1α expression on survival was investigated. Results: The strong expression of HIF-1α was observed in 16(25.4%), moderate in 14(22.2%), and weak in 15(23.8%), and no expression was found in 18(28.6%). There is statistical significance in groups(P<0.01). The expression for HIF-1α increased as pathological grade(P<0.01). The positive cells were mainly found in tumor cells infiltrating the brain at the tumor margin and necrotic zones. Every grade gliomas can express CD 34 and there is statistical significance in groups (P<0.01). the date of positive cells of CD 34 in glioma gradeⅠ, glioma gradeⅡ and glioma grade Ⅲ~Ⅳ was 35±14, 69±33, 119±49 respectively. CD 34 was significantly correlated with expression level of HIF-1α (P<0.01). The surrival time was significantly correlated with expression level of HIF-1α (P<0.01). Conclusions: The expression for CD 34 may be correlated with HIF-1α. HIF-1α is correlated with the novel ngiogenesis of the gliomas tumors. The expressions of HIF-1α and CD 34 can be used to accurately assess the prognosis in glioma patients.
