摘要
目的 观察血液回收机漂洗技术能否将血液中的肿瘤细胞去除。方法 选择繁殖能力强 ,不易受损的细胞系肠肿瘤细胞LOVO和胃肿瘤细胞SGC ,计数后分别混入浓缩的红细胞中。采用血液回收机将其过滤、离心、漂洗。在漂洗后的浓缩红细胞中分离肿瘤细胞 ,台盼蓝染色计数存活的肿瘤细胞。将分离出的肿瘤细胞培养 1 4d ,进行克隆形成实验及DNA代谢物测定 ,观察肿瘤细胞的活性。结果 经血液回收机漂洗后仍然存在肿瘤细胞 ,但活性受到显著抑制 ;细胞培养 7d时 ,未发现肿瘤细胞有克隆形成 ;培养 1 4d后 ,肿瘤细胞有克隆形成。免疫组化实验显示 ,漂洗后肿瘤细胞DNA合成能力与对照组无显著性差异。结论 单纯采用血液回收机漂洗技术可显著减少血液中的肿瘤细胞 。
Objective To observe whether cell saver could remove the tumor cells in blood. Methods RBC concentration was inoculated with intestinal cancer cells LOVO and gastric cancer cells SGC. The blood sample was centrifuged, washed through cell saver and the tumor cells were then isolated from centrifuge and the living tumor cells after being washed were cultured for two weeks. The colony forming assay was analyzed by microscope. The DNA metabolism of the tumor cells was tested. Results Tumor cells were detected after being washed through cell saver, their activity was inhibited. They could grow normally in culture after two weeks. Conclusion Washing with cell saver might reduce the tumor cells in blood but it could not delivery the tumor cells totally.
