摘要 目的 :探讨细胞凋亡及凋亡相关蛋白 Fas和 Fas L在高脂饮食肾损害中的表达。方法 :将 2 4只 Wistar大鼠随机分为对照组、肾病组及高脂饮食组 ,饲养 1 2周 ,观察血脂、尿蛋白、肾脏病理积分、肾脏细胞凋亡及 Fas和 Fas L的表达。结果 :高脂饮食组血胆固醇、甘油三酯、低密度脂蛋白、尿蛋白、肾脏病理积分高于肾病组及对照组 ,肾脏免疫组化也显示高脂饮食组细胞凋亡及 Fas和 Fas L表达增强。结论 :高脂血症可加剧肾小球损伤 ,肾脏细胞凋亡及 Fas和 Fas Objective: To determine effect of cell apoptosis and expression of Fas and FasL in glomerulosclerosis nephritic rats induced by high-lipid diet . Methods: 24 male Wistar rats were randomly divided into three groups, the control group, the nephritic group and high-lipid diet group. They were fed 12 weeks. Urinary protein excretion and serum lipid were assayed, and renal glomerulosclerosis index, renal cell apoptosis, the expression of Fas and FasL were observed . Results: The value of serum total cholesterol, trigtycerides , low-densit lipoprotein, urinary protein and renal glomerulosclerosis index in high-lipid diet group were significantly higher than those in nephritic group and control group. Renal immunohistochemistry also showed that renal cell apoptosis and the expression of Fas and FasL were also much higher in high-lipid diet group than those in nephritic group and control group. Conclusion: Our finding indicated that high-lipid diet can result in hyperlipidemia and that hyperlipidemia can lead to glomerular injury. Renal cell apoptosis, Fas and FasL may play an important role in pathogenesis of glomerulosclerosis caused by hyperlipidemia.
