摘要
[目的] 观察腹主动脉缩窄造成的压力超负荷大鼠模型左心室肌细胞B细胞淋巴瘤/白血病基因蛋白(Bcl-2蛋白)及其家族成员Bax蛋白表达的序列变化。【方法】压力超负荷大鼠各时间点(造模后1、4、7、10、21、30、40d)Bax、Bcl-2蛋白的表达用流式细胞仪进行胞内抗原定量检测。【结果】在腹主动脉缩窄之后第1天,大鼠左心室肌细胞Bax蛋白的表达率较高(63.75%),Bcl-2蛋白的表达率则较低(31.45%),Bax/Bcl-2较高(2.02);在以后的观察点中,Bax蛋白的表达率变化不大,波动于50%~60%之间,而Bcl-2蛋白的表达则于第7天升至最高值(71.75%),尔后开始下降,第30天时其表达率最低(22.8%);Bax/Bcl-2于第4天时下降,第7天时降至最低,随后开始逐渐上升,第30天达到峰值(2.4),到第40天时又下降。[结论]腹主动脉缩窄造成的压力超负荷大鼠模型左心室肌细胞凋亡发生的高峰可能在造模后的第30天。
[Objective] To explore the sequent changes of Bax and Bcl-2 protein expression in left ventricular myocytes
of pressure overloaded rats induced by abdominal aortic constriction (AAC). [Methods] Quantitative determination of
intracellular antibody was carried out by flow cytometry 1, 4, 7, 10, 14, 21, 30 and 40 days after modeling to
evaluate the protein expression of Bax and Bcl-2 after AAC. [ Results] One day after AAC, expression rate of Bax was
high (63.75% ) and that of Bcl-2 was low (31.45% ), the ratio of Bax and Bcl-2 being 2.02. Later, expression rate of
Bax stayed steady, ranging from 50% to 60%. Bcl-2 expression rate arrived the peak (71.75% ) on the 7th day after
AAC, then began to decreased and arrived the valley (22.8% ) on the 30th day after AAC. Bax and Bcl-2 ratio began to
decrease on the 4th day and was in the lowest on the 7th day, then began to increase and arrived the peak (2.4) on the
30th day and decreased on the 40th day. [ Conclusion] The maximum apoptosis in left ventricular myocytes of pressure
overloaded rats may occur on the 30th day after AAC.
