摘要 目的 :建立分离培养符合实验要求的人在位及异位子宫内膜间质细胞的方法 ,为进一步探讨子宫内膜异位症发生、发展的分子生物学机制提供一个理想的实验模型。方法 :胶原酶消化在位和异位内膜组织 ,进行原代培养 ,胰蛋白酶消化法传代培养 ,光镜、免疫细胞化学染色进行细胞鉴定。结果 :角蛋白、波形蛋白染色证实细胞纯度达 95 %以上。结论 :胶原酶消化法简便易行 ,可获得符合实验要求的人子宫内膜间质细胞 ,可以用于子宫内膜异位症发病机制。 ObjectiveTo establish the purified cell cultures of stromal cell of endometrium in vitro . MethodsCollagenase digestion,trypsase digestion,light microscope,and immunocytochemical staining were used. Endometrium and endometriotic tissues were isolated, purified,and cultured.ResultsThe purity of stromal cells was 95% identified by cytokeratin and vimentin staining. ConclusionThe purified stromal cell cultured from endometrial and endometriotic tissues are obtained by centrifugation. The cellular model will be useful to investigate the pathogenesis and therapy of endometriosis.
