摘要 根据种子生活力的氯化三苯四氮唑(TTC)染色机理,探讨了染色温度、时间、种子材料的处理对羊草(Leymuschinensis)种子胚染色效果的影响,以筛选鉴别种子生活力的最佳反应条件;并对不同基因型的羊草种子生活力进行了对比。结果表明,羊草种子生活力检验可以用如下方法,羊草去稃种子,加入磷酸缓冲液配制成0.5%的TTC溶液,在35℃恒温条件下保持反应2~3h,观察胚部染色状态,呈红颜色者为具有活力的种子,反之为失活种子。本研究建议在生产实际中应将生活力作为评价羊草种子质量的重要指标。 In order to find the optimal conditions for identifying the viability of Leymus chinensis seeds, and to compare the viability of seeds from different L. chinensis genotypes, experiments were conducted to study the staining effect of 2,3,5-triphenyltetrazolium chloride (TTC) treatment on L. chinensis seed embryos at (different) temperatures, for different time durations, and with different seed treatments. The results suggested that the viability of L. chinensis seeds can be tested for by soaking seeds with lemma and palea removed in (0.5%) TTC phosphate-buffered solution for 2-3 hours at 35℃. Seeds with embryos stained red were viable, while seeds with unstained embryos were not. It is suggested that viability is an important index for evaluating L. chinensis seed quality.
