摘要
目的:探讨使用外源性生长激素诱发继发性糖尿病的可能性及机制。方法:以28只雄性SD大鼠为实验对象,分为四组:安静组、安静注射组、运动组和运动注射组。注射重组人生长激素(recombinanthumangrowthhormone,rhGH),结合耐力训练,观察rhGH对血糖、血清胰岛素、C肽的影响,并用图像分析方法观察胰岛β细胞形态的变化。结果:①运动组的血糖值均明显低于安静组(P<0.01);而注射组与之相对应的非注射对照组相比,血糖值无显著性差异。②注射组与之相对应的非注射对照组相比,C肽显著升高(P<0.05)。③安静注射组的胰岛素水平明显高于安静对照组;运动注射组的胰岛素与单纯运动组相比,有升高的趋势。④C肽与空腹血糖的比值(C/G),运动组均明显高于安静组(P<0.001)。⑤HBCI(β细胞分泌功能指数),运动注射组比单纯运动组有明显升高(P<0.05);安静注射组与安静对照组相比,也有升高趋势。⑥β细胞占胰岛的面积比例,注射组与之相对应的非注射对照组相比,有升高的趋势;但各组之间无显著性差异。结论:rhGH可诱导大鼠产生胰岛素抵抗,rhGH可促进β细胞的胰岛素分泌。
AIM:To investigate the possibility and mechanism of the secondary diabetes induced by long time administration of ectogenesis growth hormone. METHODS:Twenty eight male SD rats were assigned into 4 groups:rest group,rest injection group,training group and training injection group.They were treated by injection of recombinant human growth hormone(rhGH) combined with endurance training.The effects of rhGH on blood glucose,serum insulin,serum C peptide were observed, and the changes of morphology of β cells was also observed with the method of image analysis. RESULTS:①The level of blood glucose in the training group was significantly lower than that in the rest group(P< 0.01), but there was no significant difference between the injection and the non injection groups.②C peptides increased significantly in the injection groups compared with the non injection groups(P< 0.05).③The serum insulin level in the rest injection group was obviously higher than that in the resting control group;that in the training injection group increased compared with that in the training group.④The ratio of C peptides to fasting glucose(C/G) in the training groups was significantly higher than that in the rest groups(P< 0.001).⑤HBCI in the training injection group increased more obviously than that in the training group;that in the rest injection group increased compared with the rest control group(P< 0.05).⑥The ratio of the areas of β cell to the pancreatic islets in the injection groups had the trend of increase,but the difference was not significant between groups.rhGH can induce insulin resistance and accelerate the insulin secretion of β cell.
