摘要
目的:为了比较针刺足阳明胃经、足少阳胆经穴对胃窦平滑肌及Oddi括约肌组织中缩胆囊素(cholecystokinin,CCK)受体基因表达的影响。方法:将60只家兔随机分为空白组、阿托品组、足三里(足阳明经)、阳陵泉(足少阳经)、四白组(足阳明经)、承筋组(足太阳经)组。除空白组外,各组均静滴阿托品造成平滑肌抑制状态,与此同时足三里、阳陵泉、四白、承筋组分别电针足三里、阳陵泉、四白及承筋穴20min。实验结束后取胃窦平滑肌及Oddi括约肌组织,采用RT-PCR方法测上述组织内缩胆囊素A受体基因(CCKA-R-mRNA)表达。结果:电针足阳明及足少阳经穴均能上调胃窦及Oddi括约肌组织中CCKA-R-mRNA的表达。对胃窦平滑肌组织CCKA-R-mRNA表达上调作用,针刺足三里(2.533±0.413)作用大于阳陵泉(1.453±0.508)及承筋组(1.098±0.230),差异有显著性意义(F=15.252,P<0.01);对奥狄氏括约肌组织CCKA-R-mRNA表达上调作用,阳陵泉组(1.921±0.655)大于承筋组(0.820±0.399),差异有显著性意义(F=6.628,P<0.01)及足三里组(1.359±0.105)。以上各组中以四白穴对上述两组织受体基因表达上调作用最强。结论:针刺足阳明、足少阳经对胃、胆运动的特异性调整作用,不仅与相关脑肠肽的释放有关,还与脑肠肽受体基因表达差异有关。
AIM:To compare the changes of cholecystokinin(CCK) receptor genes expression of gastric antrum smooth muscle and Oddi's sphincter by electro acupuncture in stomach channel of foot yangming and gall bladder channel of foot shaoyang respectively. METHODS:Sixty rabbits were randomly divided into 6 groups:blank group,atropine group,zusanli group(channel of foot yangming),yanglingquan group(channel of foot shaoyang),sibai group(channel of foot yangming) and chengjin group(channel of foot shaoyang).All groups were induced to the state of smooth muscle inhibition by intravenous drip of atropine except the blank group,meanwhile,the zusanli,yanglingquan,sibai and chengjin groups were electroacupuncted at the acupoints of zusanli,yanglingquan,sibai and chengjin for 20 minutes respectively.The tissues of gastric antrum smooth muscle and Oddi's sphincter were taken off,and the expression of CCK A receptor mRNA(CCKA R mRNA) of above tissues was detected by the reverse transcriptase polymerase chain reaction(RT PCR) method. RESULTS:The expression of CCKA R mRNA in the gastric antrum smooth muscle and Oddi's sphincter were all up regulated by electro acupuncture in the channels of foot yangming and foot shaoyang.Concerning the up regulation of expression of CCKA RmRNA in the gastric antrum smooth muscle,the effect in the zusanli group(2.533 ±0.413) was better than those in the yanglingquan(1.453±0.508) and chengjin(1.098±0.230) groups(P< 0.01).Concerning the up regulation of expression of CCKA R mRNA in Oddi's sphincter,the effect of yanglingquan group(1.921±0.655) was better than those in the chengjin group(0.820±0.399),(P< 0.01) and zusanli group(1.359±0.105).The up regulating effect on the expression of CCKA R mRNA of the two tissues in the sibai group was the best among the above groups. CONCLUSION:Electroacupuncture in the channels of foot yangming and foot shaoyang can influence the movement of stomach and gall bladder,not only related to the release of the brain gut peptide(BGP),but also related to the different genes expression of relevant BGP receptor.
