葛根素对去卵巢大鼠骨密度和骨代谢生化指标的影响 被引量:47

Effects of puerarin on the bone mineral density and biochemical markers of bone metabolism in ovariectomized rats

摘要 目的:研究葛根素对去卵巢大鼠骨密度和骨代谢生化指标的影响,探讨葛根素对雌激素缺乏引起的骨质疏松的治疗作用。方法:3月龄SD大鼠32只,随机分为单纯卵巢摘除组,假手术组,雌激素治疗组和葛根素治疗组,手术后7d,雌激素组给予苯甲酸雌二醇,200μg/kg皮下注射,每周两次;葛根素组给予葛根素注射液50mg/kgl皮下注射,1次/d;单纯卵巢摘除组与假手术组每日皮下注射与葛根素组和雌激素组同等体积的生理盐水,10周后测量大鼠骨密度、子宫系数、血尿中骨代谢生化指标。结果:与单纯卵巢摘除组相比,葛根素组大鼠骨密度(g/cm2)明显升高(0.112±0.002和0.106±0.004,t=-3.851,P<0.05),血碱性磷酸酶犤(1.26±0.14)和(2.14±0.63)mkat犦、尿钙/尿肌酐(0.209±0.069和0.237±0.050)、尿脱氧吡啶啉/尿肌酐下降(6.517±1.374和9.592±3.507),差异有显著性意义(t=3.858,-3.823,2.309,P均<0.05),血雌二醇升高犤(6.188±4.350)和(3.385±3.811)ng/L,t=3.858P<0.05犦;与雌二醇组相比,葛根素组子宫系数(g/kg)明显降低,差异有显著性意义(1.053±0.735和4.265±1.446,t=5.610,P<0.01)。结论:葛根素具有与雌激素类似的减少骨吸收,促骨形成,增加骨密度的作用,而在刺激子宫组织增生的副作用方面又远较雌激素为轻。 AIM:To investigate the effects of puerarin on the bone mineral density(BMD) and biochemical indexes of bone metabolism in ovariectomized rat,and to explore the therapeutic effect of puerarin on osteoporosis caused by the deficiency of estrogen. METHODS:Thirty two 3 month old female SD rats were randomly divided into four groups:simple ovariectomized group,sham operation group,estrogen group and puerarin group.Seven days after the ovariectomy,rats in the estrogen group were infected subcutaneously by with benzoestrofol 200 μg/kg,twice a week;rats in the puerarin group were injected subcutaneously puerarin 50 mg/kg,once a day;while rats in the simple ovariectomized group and sham operation group were injected subcutaneously with saline of the same volume as those in the estrogen and puerarin groups everyday.After 10 weeks of intervention,the BMD,index of uterus,the serum and urine biochemical markers of bone metabolism were determined. RESULTS:Compared with the simple ovariectomized group,the BMD increased obviously[(0.112±0.002)g/cm2 vs(0.106±0.004)g/cm2,t=-3.851,P< 0.05],while the serum alkaline phosphatase level[(1.26±0.14)vs (2.14±0.63)mkat],urine calcium/ urine creatinine(0.209±0.069 vs 0.237±0.050) and urine deoxypyridinoline/urine creatinine(6.517±1.374 vs 9.592±3.507) decreased significantly(t=3.858,-3.823,2.309,P < 0.05),and the level of the serum estrogen improved[(6.188±4.350)vs (3.385±3.811) ng/L,t=3.858,P< 0.05)]. Compared with the estrogen group,the index of uterus decreased significantly in the puerarin[(1.053±0.735) vs (4.265±1.446) g/kg,t =5.610,P< 0.01]. CONCLUSION:Puerarin has the same effect as estrogen in inhibiting bone resorption,inducing bone formation and increasing the bone miner density.While in the aspect of stimulating the hyperplasia of the uterus tissue,puerarin is not as obvious as estrogen.
