大鼠肝癌细胞与磷脂酰乙醇胺N-甲基转移酶2过表达细胞蛋白含量及增殖情况的比较

目的 比较大鼠肝癌细胞与磷脂酰乙醇胺N 甲基转移酶 2 (PEMT 2 )过表达细胞蛋白含量及增殖情况。方法 采用载体构建、质粒转染和鉴定方法 ,得到PEMT 2过表达细胞 ,用细胞培养、计数及考马斯亮蓝法分析大鼠肝癌细胞与PEMT 2过表达细胞蛋白含量及增殖情况。结果 PEMT 2过表达细胞的生长速度明显低于大鼠肝癌细胞 ,而胞浆蛋白及总蛋白含量却明显高于大鼠肝癌细胞 (P <0 .0 1)。结论 PEMT

Objective To compare the protein contention and proli fe ration between rat hepatoma cells and phosphatidylethanolamine-N-methyltransfe rase-2 (PEMT-2) overexpression cells.Methods Using carrier c onstruction , plasmid transfection and identificaion,the PEMT-2 overexpression cells was obtained, and protein content and proliferation of cell were analysed by ce ll culture and counts and Coomassie brilliant blue assay.Results The growth speed of PEMT-2 of overexpression cells was obviously slower than the hepatoma cells and the protein content of cytoplasm and total protein conte nt in PEMF-2 overexpression cells was obviously higher than that in the hepato ma cells(P<0.01 ).Conclusion The overexpression of PEMT-2 inhibites the development of rat hepatoma cells and makes them approach differentiation.
