摘要 目的 :研究蕈样肉芽肿 (MF)中的端粒酶与P5 3的关系 ,分析它们在MF的发病机制中的作用。方法 :使用免疫组化法检测P5 3的蛋白表达 ;聚合酶链反应 -酶联免疫吸附测定法 (PCR -ELISA)检测端粒酶。结果 :61 5 %肿瘤期MF和 2 1 4%的斑块期MF的P5 3蛋白呈阳性 ;扁平苔藓和正常皮肤均为阴性。肿瘤期MF、斑块期MF、斑片期MF、扁平苔藓和正常皮肤的端粒酶活性水平的平均A值分别为0 73 7、0 3 1 3、0 2 40、0 0 45和 0 0 44。P5 3蛋白阳性的MF的端粒酶A值为 0 745 ,P5 3阴性的MF的端粒酶A值为 0 3 2 0 ,P5 3阳性的MF的端粒酶A值明显高于P5 3阴性的MF。结论 :端粒酶活性与MF的恶性程度及分期有关 ,而且端粒酶活化可能与P5 3有关。端粒酶和P5 3均在MF的发病机制中起重要作用 ,对MF的诊断和分期有一定的意义。 Objective:To study the relationship between telomerase and P53 in patients with mycosis fungoides (MF) and analysis their roles in tumorigenesis of MF. Methods:To detect the expression of P53 protein with immunochemistry and telomerase with polymerase chain reaction-enzyme linked immunosorbent assay (PCR-ELISA). Results:The P53 protein was positive in 61.5% tumor stage of MF and 21.4% plaque stage of MF, while negative in lichen planus and normal skin. The values of telomerase activity were 0.737, 0.313, 0.240, 0.045 and 0.044 in tumor stage, plaque stage, patch stage of MF, lichen planus and normal skin respectively. In addition, the value of telomerase activity was 0.745 in MF with positive P53 protein and 0.320 in MF with negative P53 protein, the former was higher than the latter. Conclusion:Telomerase correlated with the malignant grade and stage of MF. In addition, telomerase activation was related with P53, telomerase and P53 played an important role in the pathogenesis of MF and would be useful in the diagnosis and classfication of MF.
