摘要
目的 研究氟伐他汀在高血压病人中的抗炎作用。方法 入选 12例正常人和 2 4例高血压病人。密度梯度离心法分离外周血单个核细胞 ,其分泌的白细胞介素 - 1β用ELISA法检测。随后将高血压病人分为A组和B组 ,A组用 80mg代文降压 ,B组在代文的基础上加 4 0mg的氟伐他汀。治疗 2周后再次检测外周血单个核细胞分泌的白细胞介素 - 1β。 结果 与对照组相比 ,高血压病人外周血单个核细胞血管紧张素刺激的白细胞介素 - 1β分泌增加 [(992± 15 1) pg/ml,(912± 10 2 ) pg/mlvs (5 99± 93)pg/ml,P <0 .0 5〗 ,氟伐他汀能下调血管紧张素刺激的白细胞介素 - 1β分泌 [(6 6 8± 98) pg/mlvs (92 3± 6 7)pg/ml,P <0 .0 5 ],且与降脂作用无关。结论 该研究表明高血压存在外周血单个核细胞的激活。氟伐他汀在高血压中有抗炎作用。
Objective:To study the anti-inflammatory effects of fluvastain in patients with essential hypertension.Methods: PBMCs were isolated by gradient centrifugation from 24 individuals with essential hypertension (EH) and 12 normotensive people. IL-1β concentrations in supernatant from PBMCs were measured by Enzyme-linked immunosorbent assay(ELISA).The patients with EH were then randomized to treatment with fluvastatin (40 mg/day) or matching group for 2 weeks. IL-1 β secretion by PBMCs were also measured. Results: Compared with controls, patients with EH had increased IL-1 β[ (992± 151) pg/ml,(912±102) pg/ml vs (599±93) pg/ml;P<0.05]secretion by PBMCs after stimulated by angiotensin Ⅱ. Fluvastatin treatment had a significant effect of decreasing IL-1 β[(668±98)pg/ml vs (923±67) pg/ml; P<0.05]secretion in PBMCs. The reductions were not correlated to changes in plasma lipids.Conclusions:This study shows that EH is associated with increased PBMCs activation and that treatment with fluvastatin may partly attenuate this abnormality.
