摘要
目的 进行合成肽RGDS(精氨酸 甘氨酸 天冬氨酸 丝氨酸 )抑制脑转移舌癌细胞转移机理方面的研究。方法 采用MTT法研究不同浓度的RGDS对Tb细胞粘附人工重组基底膜 (Matrigel)及纤维连接蛋白 (Fn)能力的影响 :细胞移动实验测定RGDS抑制Tb细胞在Matrigel及Fn上迁移的能力 ;明胶底物SDS PAGE电泳推测RGDS与基质金属蛋白酶的关系。结果 不同浓度的RGDS抑制了Tb细胞对Matrigel及Fn的粘附 ,并能抑制Tb细胞在Matrigel及Fn上的迁移。电泳结果表明RGDS能部分抑制由Fn诱导的基质金属蛋白酶的分泌。 结论 RGDS能抑制Tb细胞的粘附、迁移及抑制由Fn诱导的金属蛋白酶分泌 ,提示RGDS有抗肿瘤转移活性。
Objective To study the mechanism of synthetic peptide RGDS (arginine-glycin-aspartate-serine) on the metastasis of the cell line from brain metastasis of human tongue cancer. Methods MTT assay was applied to study the effects of different concentration of RGDS peptide on Tb cells' adhesive capability to Matrigel and Fibronectin. Cellular motility experiment was carried out to assay the inhibitory effect of RGDS peptide on Tb cells' motility on Matrigel and Fibronectin. The electrophoresis of substrate SDS-PAGE gels containing gelatin was used to infer the relationship between RGDS and matrix metalloproteinas. Results Different concentration of RGDS peptide showed inhibitory effect on the adhesion of Tb cells to matrigel and fibronectin. It also could restrain Tb cells from moving on matrigel and fibronectin. The result of the electrophoresis indicated that RGDS peptide could partly limit the secretion of the matrix metalloproteinas which was induced by fibronectin. Conclusion RGDS peptide has the capability to restrain Tb cells from adhering, migrating and secreting of MMPs which was induced by fibronectin. It shows that RGDS has the activity of anti-tumor metastasis.
