摘要
用随机引物法将地高辛标记克隆于PDW79和PDW75质粒中的登革Ⅱ病毒cDNA,以碱性磷酸酶偶联体系检测地高辛-cDNA及其产物,显色灵敏度近0.1pg,感染的Den·v的细胞抽提液和登革患者血清中病毒RNA液的最高稀释度分别为1/320和1/160—320。本文报告的地高辛-cDNA探针不仅快速检出登革热病毒,而且还可以作型別鉴定,是值得推荐的新方法。
Viral cDNA of the dengue virus type 2 (Den-2) cloned in the plasmid PDW79 and PDW 75 was labelled with Digoxigenin through the random primer method and used as probes of the detection of Den RNA in infected c_6/36 cells and the RNA in the serum of patient with dengue fever. The lower limit of colour development for the probe was about 0.1 pg. The detected highest dilutions of RNA extracted from infected cells and patient's serum were 1:320 and 1:160~320, respectively. Digoxigenin-labelled cDNA probes (Digc DNA probes) described in the paper was used in the detection and typical identification of dengue virus. We think that this is a new method to luorth recommend.
