摘要 目的建立酶受体分析法 (enzymereceptorassay ,ERA) ,并研究卵巢癌 3AO细胞hCG受体的结合特性。方法采用ERA ,通过竞争结合实验、受体结合的特异性实验、饱和结合实验测定 3AO细胞膜上hCG受体的特异性、饱和性 ,计算其最大结合容量 (Bmax)和亲和力 (Kd)。结果ERA测得 3AO细胞LH/hCG受体的Bmax为 14 2 .4ng/mg蛋白 ,亲和力Kd值为 16 .14 2ng/ml,未标记配体hCG抑制hCG受体对HRP—hCG特异结合的IC50 为 3.5 84 μg/ml。 结论hCG可能通过靶细胞膜上的特异性受体影响着卵巢癌的发生发展。 Objective To establish enzyme receptor assay (ERA), and to study the characteristics of hCG receptor by ERA. Methods ERA was applied to determine the maximum binding (Bmax) and affinity (Kd) of hCG receptor on 3AO cells by competitive binding tests, peculiar receptor-binding tests, and saturated binding tests. Results The Bmax and Kd values of hCG receptor were 142.4 ng/mg protein and 16. 142ng/ml, respectively. And the competitive inhibition IC50 of unlabelled hCG against HRP-hCG binding was 3. 584μg/ml. Conclusion hCG may affect the development of ovarian cancer by the peculiar receptor in 3AO cells.
