摘要
目的 :观察欧芹素乙 (imperatorin,IMP)对大鼠脑缺血再灌注损伤的作用和对缺氧 /复氧诱导人脐静脉内皮细胞ECV30 4凋亡的影响。 方法 :栓线法阻断大脑中动脉建立大鼠局灶性脑缺血再灌注损伤模型 ;以 L onga5分制评分标准对实验大鼠作行为评分 ,以图像分析仪测算实验鼠大脑梗死面积 ,细胞凋亡采用碘化丙啶染色方法 ,流式细胞术检测 ECV30 4细胞的凋亡。 结果 :IMP可显著减轻脑缺血再灌注损伤大鼠的神经损伤症状 (行为评分 ) ,缩小脑梗死面积 ;IMP可剂量依赖性降低缺氧 /复氧引起的 ECV30 4细胞的凋亡。 结论 :IMP对大鼠局灶性脑缺血再灌注损伤有明显的保护作用 ;IMP降低缺氧 /复氧引起的内皮细胞凋亡是其保护机制之一。
Objective:To study the effects of imperatorin (IMP) on rat cerebral ischemia/reperfusion injury and on apoptosis of human umbilical vein endothelial cell line ECV304 induced by hypoxia/reoxygenation. Methods: Focal cerebral ischemia/perfusion injury in rats was made by middle cerebral artery occlusion(MCAO) achieved by passing a nylon thread into the internal carotid artery. Apoptosis of ECV304 was measured by propidium iodide stain and flow cytometry. Results: IMP significantly reduced the behavior score and infarction area of cerebral ischemia/reperfusion in rats, and decreased apoptosis of ECV304 induced by hypoxia/reoxygenation in a dose dependent manner. Conclusion: IMP possesses the protective effects on rat focal cerebral ischemia/reperfusion injury, and reducing apoptosis of endothelial cells induced by hypoxia/reoxygenation is one of the mechanisms of its protective effects.
