摘要
目的 :评价吗啡抗体对吗啡依赖小鼠纳洛酮催促戒断症状的影响。方法 :采用混合酸酐法制备琥珀酰吗啡 ,并分别与蓝载体 (BC ,bluecarrier)、KLH和BSA等 3种载体交联成酯 ;常规检测抗原性 ,并免疫小鼠 ,用ELISA法检测抗体滴度 ,纳洛酮催促戒断实验来评价 3种交联物对吗啡依赖性的影响。结果 :与 3种不同载体交联的吗啡酯在 5 μg·ml- 1 包板浓度时显示了良好的抗原性 ;用其免疫的 3组小白鼠的平均抗吗啡抗体滴度都超过 1∶16 0 0 0 ;对照组小鼠及BC组、KLH组、BSA组免疫小鼠在15min内跳跃次数依次为 :2 3 6 7±s 10 2 3次 ,0 88±s 1 36次 ,2 6 3±s 4 87次 ,10 0 0±s 5 71次 ,4组小鼠跳跃次数存在显著性差异 (P <0 0 1)。结论 :与 3种载体交联的吗啡酯都能诱导产生高滴度的抗体 ,对吗啡诱导的成瘾性抑制作用有所不同 ,其中BC偶联的吗啡酯较其它两种载体 -吗啡酯交联获得更好的效果。
Objective : To evaluate the effect of morphine antibody on withdrawal symptoms. Methods : The conjugates of morphine with BSA, KLH and blue carrier as carrier protein were prepared by mixed anhydride method, and their antigenicity was detected before immunization. The titres of antibody from mice immunized with three kinds of conjugates were determined respectively by ELISA. The withdrawal symptoms were induced by the opioid receptor antagonist naloxone to assess morphine dependence. Results : Three morphine conjugates showed good antigenicity in 5 μg·ml -1 coating concentration. The average titres of the three groups were over 1∶16 000. The number of skipping in the control, BC, KLH and BSA groups was 23 67± s 10 23,0 88± s 1 36,2 63± s 4 87 and 10 00± s 5 71 times in 15 minutes, respectively, showing significant difference ( P <0 01). Conclusion : Morphine-carrier conjugates can elicit high-titres antibody responses, and an inhibition on addiction of mice can be induced by morphine. Among three kinds of morphine conjugates, morphine-blue carrier shows a more effective, long-lasting anti-morphine immune response.
