摘要 目的 :观察神经肽Y(NPY)诱导大鼠心肌细胞肥大效应。方法 :用不同浓度NPY(10nmol/L、10 0nmol/L)刺激心肌细胞 ,应用3 H Leu掺入量法测定心肌细胞蛋白质合成速率、RT PCR法测心肌细胞c junmRNA表达。结果 :1.心肌细胞3 H Leu掺入量测定 :较大剂量NPY刺激心肌细胞蛋白合成速率显著增加。与对照组相比 ,NPY10nmol/L组3 H Leu掺入量有所增高 ,但差异无显著性 ,而NPY10 0nmol/L组心肌细胞 3H Leu掺入量较对照组明显增高 (P <0 .0 5 )。 2 .心肌细胞内c junmRNA表达 :NPY组心肌细胞c junmRNA的RT PCR产物量明显高于对照组(P <0 .0 0 1)。结论 :NPY刺激心肌细胞蛋白质合成增加、心肌细胞原癌基因 (肥大早期反应基因 )c junmRNA表达增加 ,提示NPY可通过其生长因子样效应 ,诱导心肌细胞肥大。 Objective:To investigate the effect of neuropeptide Y(NPY) on cardiomyocytes hypertrophy in rats.Method: Cardiomyocytes of neonatal Wistar rats were cultured with NPY in various concentrations (10 nmol/L?100 nmol/L). To assess protein synthesis rate and c jun mRNA expression in cardiomyocytes, 3H Leu incorporation and RT PCR were used. Result: 1. 3H Leu incorporation in cardiomyocytes:Stimulated with larger dosage of NPY,the protein synthesis rate in cardiomyocytes was elevated. Although 3H Leu incorporation in NPY(10 nmol/L) group was higher than in control group,there were no significant difference between two groups;Compared with control group, 3H Leu incorporation in NPY(100 nmol/L) group were increased significantly( P< 0.05). 2. c jun mRNA expression in cardiomyocytes: RT PCR production of c jun mRNA in NPY group was enhanced considerably compared with control group( P< 0.001).Conclusion:The enhancement of protein synthesis rate and oncogene c jun (hypertrophic IE gene) mRNA expression due to simulation of NPY suggests that NPY induces hypertrophy of cardiomyocytes like growth factor.
