中药改善孕期巨细胞病毒感染对子代生长发育影响的动物实验研究 被引量:6

Improving the Growth and Development after Cytomegalovirus Infection in Early Pregnancy with Traditional Chinese Medicine-Jinyebaidu in the Guinea Pig Offspring Model

摘要 目的 :探讨中药金叶败毒改善孕早期豚鼠感染GPCMV所导致的子代生长发育异常的效果。方法 :运用N PCRC ,筛选无感染史的孕早期豚鼠随机分为 3组 (每组 2 5只 ) :空白对照组无特殊处理 ,阳性对照组腹腔接种病毒 ,观察组接种病毒并给予中药金叶败毒 ,孕中期处死豚鼠 ,观察金叶败毒改善子代生长发育的效果。结果 :阳性对照组和观察组的妊娠豚鼠感染率分别为 10 0 % (2 5 /2 5 )和 4 4 % (11/2 5 )、胎盘感染率分别为 10 0 % (2 3/2 3)和6 6 6 7% (12 /18)、胎仔感染率分别为 95 .5 6 % (4 3/45 )和 4 7.5 4 % (2 9/6 1)、死胎率分别为 31.11% (14 /45 )和 13 11%(8/6 1) ,差异均具有统计学意义 (χ2 =39.4 4P =0 .0 0 1,χ2 =8.98P =0 .0 0 3,χ2 =2 7.4 0P =0 0 0 1,χ2 =5 .10P =0 .0 2 4 )。结论 :金叶败毒可显著降低孕期GPCMV感染所导致的死胎 ,其作用机制可能与减少胎盘感染有关。 Objective:To discuss the effect of improving the growth and development in guinea pig offspring after cytomegalovirus (GPCMV) infection in early pregnancy with Traditional Chinese Medicine Jinyebaidu. Methods:The prospectively randomized controlled trials were done.The sexually mature guinea pigs were scanned for negative GPCMV DNA using Nested PCR. Put the male and female ones in the same cage.Total 75 pregnant guinea pigs in the first trimester were studied:25 of them treated nothing specially,other 25 only inoculated suspension of GPMCV intraperitoneal,anotherly 25 took Jinyebaidu through stomach perfusionafter inoculation.The pups growth and development was followed up. Results:Jinyebaidu can significantly lower the GPCMV maternal infection rate from 100%(25/25)to 44%(11/25) (χ 2=39.44 P= 0.001),the placental infection rate from 100%(23/23)to 66 67%(12/18) (χ 2=8.98 P=0.003) ,the congenital infection rate from 95.56%(43/45)to 47.54%(29/61) ( χ 2=27.40 P=0.001) ,and the stillborn rate from 31.11%(14/45)to 13.11%(8/61)( χ 2=5.10 P=0.024). Conclusions:Traditional Chinese Medicine Jinyebaidu markedly reduces the stillborn rate after GPCMV maternal infection.Decreasing the placental infection rate might be one of its mechanism.
