摘要
目的 :为了定量检测系统性红斑狼疮 (systemiclupuserythematosus,SLE)病人血清中各种抗可提取核抗原 (extractednuclearantigens ,ENA)自身抗体的滴度 ,并探讨其临床意义。方法 :用自制的抗各种ENA(包括 :Sm、RNP、ssA、ssB、scl 70、和Jo 1)的单克隆抗体包被ELISA板和小牛胸腺丙酮粉作为ENA抗原以夹心ELISA法定量检测 31例SLE病人、5 0例非结缔组织病 (non connectivetissuedisease ,non CTD)病人和 6 0例健康人血清中的各种抗ENA自身抗体的滴度。并以健康人血清中各ENA抗体的滴度的均数加上 3倍标准差 ( x +3s)的滴度为标准 ,以判定各种ENA抗体的检出阴、阳性。结果 :各种ENA抗体在SLE病人中的滴度和阳性率分别是 :Anti Sm为 75 8±4 0 3单位 (arbitraryunits,Au)和 38.7%、Anti u1 RNP为 818± 4 17Au和 5 5 %、Anti ssA为 6 15± 318Au和 19.4 %、Anti ssB为 5 6 5± 2 78Au和 16 .1%、Anti Scl 70为 2 30± 95Au和 3.2 %、Anti Jo 1为 2 71± 89Au和 3.2 %。其中 ,Anti Sm、u1 RNP、ssA和ssB四种抗体在SLE病人中的血清滴度和阳性率都明显高于non CTD和健康人 (P <0 .0 1)。结论 :本实验方法能定量检测SLE病人血清中各种ENA抗体滴度 ,并对SLE的诊断具有一定参考价值。
Objective:To analyse quantitatively the various anti ENA(extracted nuclear antigens) autoantibodies in the systemic lupus erythematosus(SLE) and study the clinical significance of these antibodies in SLE.Methods:the levels of these autoantibodies,including anti Sm,u1 RNP,ssA,ssB,Scl 70,and Jo 1,in the serum from 31 SLE patients,50 non CTD patients and 60 healthy people,were detected by a quantitative sandwich ELISA kit using monoclonal antibodies to Sm,u1 RNP,ssA,ssB,Scl 70,and Jo 1.The prevalence of these antibodies were also detected by this ELISA kit according to its cut off value(the titer of the means plus 3 standard deviation of the 60 healthy people).Results:The level and positive rate of anti Sm,u1 RNP,ssA,and ssB antidodies were 758±403 arbitrary units(AU)and 38.7%,818±417 AU and 55%,615±318 AU and 19.4%,and 565±278 AU and 16.1% respectively in SLE patients,significantly higher than that of non CTD patients and healthy people(p<0.01),Conclusion:The results indicated that the various anti ENA autoantibodies in the SLE could be quantitatively detected by the quantitative sandwich ELISA kit and the detection of these anti ENA autoantibodies is useful for diagnosis in patients with SLE.
