鼻咽癌p53基因的表达及功能状态研究

摘要 目的:研究鼻咽癌(NPC)中p53表达及功能。方法:对38例N:PC和16例对照样本中的p53、p21 mRNA进行RT—PCR定量分析。结果:(1)NPC组p53mRNA的水平较高,差异有统计学意义(t=2.14,P<0.05);p21 mRNA的水平两组相同。(2)在相关分析中,对照组p21mRNA水平随p53 mRNA的水平升高而升高,相关具有高度显著性,[(γ p53一p21)=0.654,P<O.01]。在NPC组虽然也观察到类似的相关关系,但γ值比对照组小。(3)回归分析表明,NPC组p53一p21直线回归方程的斜率,明显小于对照组的斜率。结论:NPC中p53表达增加而功能异常。 Objective: Studying the expression and function status of p53 . Methods: RT-PCR quantitative analysis and RT-SSCP analysis of p53 and p21 mRNA were used in 38 NPC and 16 control samples. Results: 1. The level of p53 mRNA is higher in NPC group than that in controls, the difference is statistically significant (t = 2.14, P < 0.05). The level of p21 mRNA is same between NPCs and controls. 2. In correlation analysis, the quantities of p21 mRNA in controls increase with the increase of p53 mRNA expression, there is an apparent correlation [ γ (p53 - p2l) = 0.654, P < 0.01]. Although similar correlation was observed in NPCs, the correlation coefficients is much smaller. 3. Regression analysis reveals that the slope of linear regression equation of p53 - p21 in NPCs is (0.4958), apparently smaller than 0.9235 of control. Conclusion: p53 overexpression but inactivation in NPC.
