胚胎干细胞向神经细胞定向诱导分化的研究 被引量:6

摘要 目的 探讨由胚胎干细胞向神经细胞方向分化的最佳条件。 方法 从小鼠胚泡内细胞团中获取胚胎干细胞进行体外培养 ,向培养液中分别加入维甲酸、大脑皮层细胞条件培养液 +维甲酸、β 巯基乙醇 +维甲酸+大脑皮层细胞条件培养液 ,通过形态学观察和神经元特异性烯醇化酶 (NSE)免疫细胞化学染色对分化细胞进行鉴定。 结果 用维甲酸、β 巯基乙醇和大脑皮层细胞条件培养液联合诱导胚胎干细胞 ,可使 70 %以上的胚胎干细胞分化为NSE免疫阳性的神经细胞。 结论 维甲酸、β Objective To explore the optimal condition on which embryonic stem cell may be induced to differentiate directly into nerve cell. Methods Embryonic stem cells derived from inner cell masses of mouse balastocysts were cultured in the medium with RA,RA+conditioned medium of cerebral cortex and β ME+RA+ conditioned medium of cerebral cortex respectively.Differentiated cells were determined by morphological observation and NSE immunocytochemical staining. Result The result showed that over 70 percentage of differentiated cells were nerve cells which were NSE immunopositive.Conclusion\ It suggests that the culture system with RA,β ME and conditioned medium of cerebral cortex is an ideal inductive microenviorment,and moreover,these three fractors are intercomplementary and coodinative with each other for inducing ES cells to differentiating directly into nerve cells.\;[
