化疗药物诱导宫颈癌Hela细胞凋亡的研究 被引量:1

摘要 目的观察顺铂(CDDP)、长春新碱(VCR)、平阳霉素(BLM)、顺铂+平阳霉素、顺铂+长春新碱在体外对人宫颈肿瘤细胞的作用。方法用四甲基偶氮唑盐(MTT)测定法观察药物对Hela细胞的影响,用流式细胞仪观察细胞凋亡变化。结果CDDP、VCR、BLM单用对Hela细胞抑制率和凋亡率均高于联合用药组。各用药组对细胞抑制率均高于凋亡率,仅CDDP组两作用相近。结论CDDP、VCR、BLM能有效抑制Hela细胞生长,联合应用出现拮抗作用。 Objective To study the cytotoxic effects of chemotherapeutic agents on human cervical cancer line Hela cell in vitro.Methods The cytotoxicity on human cervical cancer cell line Hela in vitro was determined in MTT assay,and the apoptosis was determined by flow cytometry.Results Inhibition rate and apoptosis index produced by separate use of cisplatin(CDDP),vincristine(VCR),or pingyangmycin(BLM)all got higher values than the combination use did;inhibi-tion rates were all higher than apoptosis rates among the test groups,but CDDP group got two similar values.Conclusion CDDP,VCR and BLM can effectively inhibit Hela cell's growth,but their combination use may make antagonist action.
