兔急性缺血-再灌注损伤模型下玻璃体内的谷氨酸浓度变化

摘要 为研究在急性缺血 -再灌注损伤模型下玻璃体内的谷氨酸浓度变化 ,将 3 6只雄性大耳白 ,分为 6组 ,每组 6只。 1~ 4组为实验组 ,在结扎睫状动脉、眼外肌、视神经 45分钟后分别恢复再灌注 0、15、45、75分钟 ,以右眼作为实验眼 ;空白对照组 6只直接处死动物后取眼球 ;实验对照组只做球结膜切开术然后取眼球。利用 6-氨基喹啉 -N-琥珀酰亚胺基氨基甲酸酯柱前衍生 (AQC) ,Waters高效液相色谱仪测得玻璃体内的谷氨酸含量。结果显示 ,实验组中 0分钟、45分钟 (P<0 .0 1)以及 15分钟 (P<0 .0 5 )的玻璃体内谷氨酸较实验对照组和空白对照组水平明显增高 ,75分钟组 (P=0 .71)和实验对照组 (P=0 .963 )没有显著差异。提示急性缺血 -再灌注可导致玻璃体内的谷氨酸水平增高 ,其兴奋性毒性可能参与了急性缺血 -再灌注对视网膜的损伤。
