外源性p53诱导肝癌细胞Wnt通路抑制因子Dickkopf-1的表达 被引量:2

Ectogenetic p53 May Promote mRNA Expression of Wnt Signaling Inhibitor Dickkopf-1 in Hep3B Hepatoma Cells

摘要 研究p53对Wnt通路抑制抑制因子Dickkopf-1(DKK-1)表达的调节作用。将携带p53基因的复制缺陷型腺病毒栽体(Adp53)导入到p53缺失的人肝癌细胞株Hep3B中,以RT-PCR技术检测p53对DKK-1表达的调节作用。检测结果表明DKK-1mRNA水平在转染p53 20 h后即有明显升高,其中以32 h达最高水平,随后逐渐降低。量效关系研究表明在转染剂量为0.5、5、50 pfu/cell的Adp53时DKK-1mRNA表达均有显著增高,尤以50 pfu/cell时表达水平最高。提示p53能明显诱导Wnt通路抑制因子DKK-1的mRNA表达。 To examine whether p53 has any regulatory effects on the expression of Dickkopf-1 ( Dkk-1), a Wnt pathway inhibitor, Hep3B hepatoma cells, were transfected with areplication-defective adenovirus encoding p53(Adp53), and then dickkopf-1 (DKK-1) mRNA expression was detected by RT-PCR. Dkk-1 mRNA expression in Hep3B cells was initially potentiated by transfection with Adp53 after 20 h. A peak expression was seen at 32 h and the expression declined afterwards. Dose-effect studies revealed that Adp53 in the range of 0. 5, 5 and 50 pfu/cell could promote the expression of DKK-1 mRNA, with the highest expression at the dose of 50 pfu/cell. Ectogenetic p53 promoted mRNA expression of Wnt signaling pathway inhibitor dickkopf-1 in Hep3B hepatoma cells.
