摘要
目的 :建立脑膜瘤短期原代培养的体外模型 ,研究羟基脲对脑膜瘤培养细胞的抑制作用及其可能机理。方法 :进行脑膜瘤细胞原代培养 ,应用羟基脲作药敏试验 ,利用细胞计数、MTT法、流式细胞仪等检测结果。结果 :显微镜下可见羟基脲明显影响脑膜瘤培养细胞的生长状态 ;5× 10 -3 mol/L组的细胞数减少 84 % ,5× 10 -4mol/L组减少 4 6 % ,而 5× 10 -5mol/L组减少 2 0 % (P<0 .0 1) ;5× 10 -3 mol/L组的细胞活力减少 86 % ,5× 10 -4mol/L组减少 6 0 % ,而 5× 10 -5mol/L组减少 2 8% (P<0 .0 1)。应用流式细胞仪检测细胞凋亡 ,5× 10 -3 mol/L组的细胞凋亡率增加了 8.82倍 ,5× 10 -4mol/L组增加了 3.94倍 ,而 5× 10 -5mol/L组增加了 0 .73倍 (P<0 .0 1)。结论 :羟基脲能抑制脑膜瘤培养细胞的增生 ,且呈浓度依赖性 ;羟基脲抑制脑膜瘤培养细胞的增生可能与羟基脲诱导其凋亡有关。
Objective: To investigate the tumor-suppression effect of hydroxyurea on meningioma cells and its possible mechanism. Methods: The meningioma cells were cultured in medium containing varied doses of hydroxyurea (5×10 -3mol/L, 5×10 -4mol/L, 5×10 -5mol/L), the cell growth was measured by MTT method, cell apoptosis was observed with flow cytometry (FCM). Results: MTT measurement demonstrated that the administration of hydroxyurea led to a dose-dependent suppression in cell proliferation and FCM showed a dose-dependent increase in apoptosis rate. Conclusion: Hydroxyurea can inhibit meningioma cell growth in vitro, which is most likely associated with apoptosis of the tumor cells.
