摘要
目的 :探讨外源性 FHIT基因的表达对胃癌细胞株凋亡作用的影响。方法 :将载有人外源性 FHIT基因的真核表达质粒 p Rc CMV- FHIT用脂质体介导转入胃癌细胞株 MGC- 80 3,用流式细胞仪、普通光镜、透射电镜观察 FHIT基因的表达对 MGC- 80 3细胞凋亡的影响。结果 :转染 FHIT基因的 MGC- 80 3细胞的凋亡水平 (32 % )与空质粒转染组 (12 .15 % )相比明显增高 ,且存在显著性差异 (P<0 .0 1) ,同时细胞的生长周期出现了明显的 G0 /G1期阻滞 (6 4.375 % vs 4 9.5 % )。结论 :外源性 FHIT基因能诱导胃癌细胞株 MGC- 80 3凋亡 ,细胞生长周期阻滞。
Objective: To investigate the effect of fragile hisdidine triad (FHIT) gene on the apoptosis of gastric cancer cells. Methods: FHIT gene was transfected into gastric cancer cell line MGC-803 by liposome. Western blot and immunohistochemical assay were employed to detect the expression of FHIT. Apoptosis and cell cycle were analyzed by flow cytometry (FCM). Morphological changes were observed by light and electron microscope. Results: Stable expression of FHIT protein in transfected MGC-803 cells was obtained. The rates of G0/G1 phase and apoptosis cells were significantly higher in FHIT-transfected MGC-803 cells than those in control cells (64.4% vs 49.5% and 32.0% vs 12.2%, respectively). Conclusion: The results indicate that FHIT gene might be involved in the apoptosis of gastric cancer cells.
