摘要
利用大肠杆菌高效表达系统表达大豆Kunitz型胰蛋白酶抑制剂 (SBTi A2 )Tid,并利用亲和层析的方法纯化得到有抑制胰蛋白酶活性的SBTid.对重组SBTid的 pH稳定性和温度稳定性与天然SBTia进行了比较研究 .结果表明重组SBTid在高 pH下具有更好的稳定性 。
One kind of Soybean kuntiz-type trypsin inhibitors(SBTi)-Ti^(d) was expressed in E.coli and purified by affinity chromatograph. The recombinant Ti^(d) showed trypsin inhibitory activity in vitro . The pH stability and thermal stability of Ti^(a) and recombinant Ti^(d) were studied. Compared with the natural SBTi^(a),SBTi^(d) exhibited a better stability in a high pH spectrum and a similar thermal stability at less than 50 ℃ .These data suggested that SBTi^(d) has a promising application potential as a bio-pesticide and insect-resistant gene in crop designing.
