摘要
以无菌种子萌发的子叶、茎尖和茎段及当年生半木质化带腋芽茎段为外植体,MS为基本培养基,附加BA0.5~1.0mg/L+NAA0.1mg/L的培养基,有利于子叶及茎尖的诱导培养,诱导率可达85%,附加BA2.0~3.0mg/L+NAA0.2mg/L时,则有利于带腋芽茎段的诱导培养,诱导率80%;附加BA2.0mg/L+2,4-D0.2~0.4mg/L时,有利于不定芽的分化和增殖生长,月增殖系数为4.0;生根适宜的培养基为1/2MS+NAA0.2mg/L,生根率达75%。
Cotyledons, shoot tips, stem sections and semi-lignified stem sections with axillary buds derived from sterilized seeds of Acacia dealbata were used as explants in tissue culture. The optimum media was MS + BA 0.5-1.0 mg/L + NAA 0.1 mg/L for the induction culture of cotyledons and shoot tips (with an induction rate of 85%), MS + BA 2.0-3.0 mg/L + NAA 0.2 mg/L for the induction culture of stem sections with axillary buds (with an induction rate of 80%), MS + BA 2.0 mg/L + 2,4-D 0.2-0.4 mg/L for the differentiation of adventitious buds and for proliferation growth (with a monthly proliferation coefficient of 4.0), and 1/2MS + NAA 0.2 mg/L for rooting, with a rooting rate of 75%.
