摘要
应用DAS-ELISA,RT-PCR和IC-RT-PCR技术检测百合无症病毒(LSV)。结果表明:RT-PCR和IC-RT-PCR分别可从2ng和200ng百合叶片组织中检测出LSV,分别是DAS-ELISA敏感度的1000倍和10倍。百合鳞茎的不同取样部位对PCR检测结果的影响较大,RT-PCR难以从鳞片和根部检测出病毒,但IC-RT-PCR可以检测出病毒。
Lily symptomless virus(LSV) was detected by DAS-ELISA, RT-PCR and IC-RT-PCR methods. The results showed that LSV was detected from dilutions equivalent to 2?ng and 200?ng of lily leaf materials by RT-PCR and IC-RT-PCR, respectively. RT-PCR and IC-RT-PCR were 1?000-fold and 10-fold more sensitive than DAS-ELISA. The PCR result was affected by different sampling parts of lily bulb and RT-PCR cannot detect LSV from scale and root, but IC-RT-PCR can detect.
