摘要
目的:探讨肝素酶反义寡核苷营酸对肝毒酶mRNA表达的影响。方法:食管癌EC9706细胞分6组培养,5组分别用设计合成的4条封闭肝素酶不同基因位点的反义寡核苷酸(ASODN1、ASODN2、ASODN3、ASODN4)及1条无关寡核苷酸(N-ODN)转染,另1组不转染。采用完整细胞原位杂交技术观察各组EC9706细胞中肝素酶mR-NA表达情况。结果:4条肝素酶反义寡核苷酸转染的EC9706细胞中肝素酶mRNA表达差异无统计学意义(P>0.05),但均显著低于无关寡核苷酸组(P<0.05)。结论:肝素酶反义寡核苷酸序列可抑制食管癌EC9706细胞中肝素酶mRNA表达。
Aim : To investigate the effects of antisense oligodexynucleotide (ASODN) on mRNA expression of hepara-nase in human esophageal cancer EC9706 cells. Methods: One non-sense oligodexynucletide (N-ODN) and five ASODNs against different heparanase mRNA sites were transfected into EC9706 cells, then the expression of heparanase mRNA in EC9706 cells was studied by in situ hybridization. Results: The expression of heparanase mRNA could be inhibited by ASODNs. There was no significant difference among five ASODNs (P >0.05) ,but there was significant difference between ASODNs and N-ODN (P<0.05). Conclusion: The expression of heparanase mRNA in EC9706 cells could be inhibited by ASODNs.
