摘要
目的:检测神经母细胞瘤(NB)组织中TRAIL、Caspase-3的表达。方法:运用原位杂交、免疫组化及TUNEL法检测TRAIL、Caspase-3在26例NB、9例神经节细胞瘤和10例正常肾上腺组织中的表达及细胞凋亡。结果:①TRAIL、Caspase-3及凋亡指数(AI)在NB、神经节细胞瘤及正常组织中的表达差异无统计学意义(P均>0.05)。②NB中TRAIL的表达与年龄、性别、部位、组织类型、INSS分期等无关(P>0.05),Caspase-3、AI的表达与组织类型、临床分期、病理分型有关(P<0.05)。③3种组织中Caspase-3和AI呈正相关(在NB、神经节细胞瘤和正常肾上腺组织中r分别=0.888,0.989,0.958,P均=0.000);此外,在7例Caspase-3(+)表达的NB中,TRAIL和Caspase-3呈正相关(r=0.974,P=0.001)。结论:TRAIL通过激活Caspase-3可以显著诱导NB细胞凋亡,有助于抑制NB的生长。
Aim: To investigate the faction and clinic value of TRAIL and Caspase-3 in neuroblastoma. Methods: The in-situ hybrization technique, immunohistochemistory S-P method and TUNEL method were adopted for detection of expression of TRAIL mRNA, Caspase-3 protein and apoptotic cell. ReSUltS:①The expressions of TRAIL, Caspase-3 and apoptosis index indicated no statistical difference ( P > 0. 05 ) among neuroblastoma, ganglioneuroblastoma and normal ad-renmedullary tissue.②No difference of TRAIL expression was found in groups of neuroblastoma ( P>0.05) , but expression of Caspase-3 and apoptosis index were related with tissue type , clinical stage and histological classification. ( P < 0.05). ③No relationship was found between TRAIL and Caspase-3 in all of three kinds of tissues; but expression of TRAIL was irrelevant to expression of apoptosis index; and Caspase-3 and AI manifested positive relation ( r = 0. 888, 0.989,0.958, P =0.000). Moreover, TRAIL and Caspase-3 manifested positive relation in seven cases neuroblastoma of Caspase-3 ( + ) (r =0.974, P =0.001 ). Conclusion: TRAIL can markedly induce neuroblastoma cell apoptosis which primarily pass to activate Caspase-3 , and which is conductive to repress growth of neuroblastoma.
