摘要
在pH 6 .8的磷酸盐缓冲溶液中 ,血清白蛋白与铜 乙酰丙酮络合物反应作用生成一种在汞电极上吸附性很弱的铜 血清白蛋白惰性络合物 ,使铜 乙酰丙酮络合物在 - 0 .2 8V(vs .SCE)络合吸附波还原峰电流降低 ,电流降低值与 1~ 30mg/L范围内牛血清白蛋白 (BSA)或 1~ 32mg/L范围内人血清白蛋白 (HSA)呈线性关系 ;检出限分别为 0 .5和 0 .6mg/L。运用该法测定了人血清样品中蛋白质含量 。
The interaction of protein with Cu(II)-acetylacetone complex in pH 6.8 NaH2PO4-Na2HPO4 buffer solution has been studied by single sweep polarography. Adding bovine serum albumin (BSA)/human serum albumin (HSA) into the Cu(II)-acetylacetone complex solution results in the formation of an electrochemical non-active complex of Cu(II)- BSA/HSA with much less adsorption than Cu(II)-acetylacetone complex at the mercury electrode, and causes the current decrease of the adsorptive peak of Cu(II)-acetylacetone complex. The Deltai(p) is proportional to the protein concentration from 1 mg/L to 30 mg/L for BSA and from 1 mg/L to 32 mg/L for HSA. The detection limit is 0.5 mg/L for BSA and 0.6 mg/L for HSA. The method was used for the determination of total protein in human serum sample. The result is satisfactory.
