Determination of Ciprofloxacin in Plasma by Micelle Capillary Electrophoresis On-line Sweeping Concentration Technique

摘要 采用在线Sweeping(推扫 )富集技术 ,建立了胶束毛细管电泳法测定血液中痕量乳酸环丙沙星的方法。考察了背景溶液pH值、SDS浓度、进样时间、血样预处理方法等对乳酸环丙沙星富集效果的影响。使用未涂层的毛细管柱 (5 5cm× 75 μmi.d .,有效柱长 4 7cm) ,30mmol/L硼砂 +80mmol/L十二烷基硫酸钠 (pH =9.4 0 )为背景溶液 ,在紫外检测波长 2 5 4nm、运行电压 1 8kV条件下 ,血浆样品用乙腈除蛋白后直接在线Sweep ing富集 ,富集倍数可达 6 0 0倍。线性范围在 0 .0 4~ 1 0mg/L (r =0 .999,n =8)。检出限为 0 .0 1mg/L。本方法减少了样品预处理的繁琐过程 ,弥补了毛细管电泳 (CE)在测定血液中痕量组分方面的不足 。 A method for the electrmination of trace ciprofloxacin in plasma was established by the micelle capillary electrophoresis on-line sweeping enrichment technique. The effect of pH in background solution, the concentration of sodium dodecyl sulfate (SDS), injection time, plasma sample pretreatment method on the enrichment of ciprofloxacin was investigated. The experiments were carried out in an uncoated fused silica capillary (55 cm x 75 mum i.d., effective length 47 cm) under the following conditions: background solution of 30 mmol/L borate and 80 mmol/L SDS (pH 9.40); detection wavelength 254 nm; run voltage 18 kV. The plasma sample was directly injected into the column after it was simply deproteinized with acetonitrile. Up to 600-fold concentration of the ciprofloxacin can be enriched. The good linear relation was obtained (r = 0.999, n = 8) in range of 0.04 similar to 10 mg/L. The detection limit was 0.01 mg/L, this method has been applied successfully to determine trace drugs in clinical analysis.
