阿维链霉菌bkdF的基因中断对阿维菌素合成的影响 被引量:3

摘要 以阿维菌素B组分菌株StreptomycesavermitilisBjbm0 0 0 6为出发菌株 ,用PCR方法构建支链α 酮酸脱氢酶基因bkdF(Branched chainα ketoaciddehydrogenasegene)的重组质粒pHJ5 816 (pHZ135 8 bkdF&Ermr)对其进行基因中断 ,得到重组菌株Bjbm5 816。经HPLC检测和核磁共振分析发现 ,Bjbm5 816发酵产物产生的单一组分新化合物为Oligo mycinA。 Bjbm0006 was a strain which produces four avermectin B component. bkdF (branched-chain α-keto acid dehydrogenase gene) gene involved in the biosynthesis of avermectin B was cloned by PCR from Bjbm0006 chromosome, and inserted with a Erm r gene to construct a recombinant plasmids pHJ5816 (pHZ1358∷bkdF&Erm r). The bkdF-interrupted mutant Bjbm5816 was received through homologous double crossover between the pHJ5816 and the chromosome. The interruptant Bjbm5816 produce single novel compound and it was attested to be Oligomycin A by MS, 1H-NMR, 13 C-NMR and two-dimension NMR.
