组织培养对乳胰B细胞功能和形态的影响

Effects of culture on activities of islets of neonatal pancreases

本文报道了不同培养时间对乳胰组织培养后胰岛活性的影响。通过对各组培养液胰岛素含量的测定和使用免疫组化与显微图象分析技术对 B 细胞内胰岛素的定性和定量观察表明,新鲜乳胰碎片行短期组织培养(2～8天),可获得良好的效果。

In order to study the effects of cu-lture on the activities of islets,120pancreases were divided into five groups:group Ⅰ,fresh fragments of pancreases;group Ⅱ to Ⅴ,fragments of pancrea-ses cultured at 2,4,8 or 12 days res-pectively.The insulin content releasedin the medium during culture,the nu-mber of insulin-containing cells(B ce-lls)in the sections of immunogold-silver staining(IGSS)and the contentof immunogold-silver(IGS)in B ce-lls in group Ⅴ were significantly lessthan that of the other cultured groupsand the fresh group,but no differencewas found between the other cult-ured groups and the fresh group.Th-ese results showed that fresh fragme-nts of neonatal pancreas cultured forshort term may perfectly maintain theactivities of islets.
