血浆中麝香酮的气相色谱测定方法学研究

STUDY ON QUANTITATIVE IN DETERMINATION OF MUSCONE IN PLASMA BY GAS CHROMATOGRAPHY

本文建立了用气相色谱测定麝香酮血药浓度的方法。以高纯氮为载气,OV—1交联甲基硅烷毛细管柱,FID检测,正十九烷为内标,进行定量测定。检测限为0.8ng(S/N=2),最低检测浓度为50μg/l血浆,麝香酮血药浓度在1.25～25.00μg/l范围内线性关系良好(r=0.9999),回收率为98.5±4.8%。该方法重视性好,特异性强。用本法测定了大鼠口服给药的药代动力学参数。

The GC method was established under the following conditions for the separationand quantitative determination of muscone in plasma: high purity N_2 as the carrier gas, OV-1crosslinked methylsilicone column as the capillary column, FID as the detector, n-Nonadecane as theinternal standard. A linear calibration curve was obtatined by plotting concentration vs peak area ratio,and the linear relatiohship was fine in the range of 1.25 mg/l plasma (r=0.9999). The average re-covery was 98.5±4.8%. The minimum detectable quantity of muscone was 0.8ng/2μl (squivalentto 50 μg/l plasa). The method is simple,sensitive,specific, reproduceable and suitable for the pharma-cokinetic study of muscone. With regard to oral administration in the dosage of 80 mg/kg to rats, theT1/2Ka was 22.0 min,t_(max) was 74.4min,C_(max)was 1.44mg/l, t1/2β was 196.1min demonstrating thatthe absorption and elimination of muscone after p. o. administration were rapid.
