胃、直结肠癌病人口服优福定和复方喃氟啶胶囊后5-氟尿嘧啶血药浓度的测定

A COMPARATIVE STUDY OF 5-FLUOROURACIL BLOOD CONCENTRATION AFTER ORAL ADMINISTRATION OF UFT AND FT207CO IN PATIENTS WITH GASTRIC AND COLORECTAL CANCER

我们于1988年1月—1991年12月采用高效液相色谱法(HPLC)对138例胃、直结肠癌患者在口服优福定片剂(UFT)或复方喃氟啶胶囊(FT207CO),各含相当于喃氟啶(FT207)200mg后进行5-氟尿嘧啶(5-FU)的血浓度测定。在口服优福定71例中,胃癌组38例在口服后0.5,1.0,和2.0h的5-氟尿嘧啶平均血浓度(mg/l ?±SD)分别为6.69±8.72(n=28),4.34±3.84(n=32)和5.76±18.12(n=24),直结肠癌组33例分别为4.25±4.34(n=24),4.50±5.62(n=27)和2.39±2.78(n=28);在口服复方喃氟啶胶囊67例中,胃癌组36例分别为4.89±5.09(n=31),3.92±3.50(n=33)和2.59±4.35(n=31),直结肠癌组31例分别为2.97±3.55(n=28),3.32±3.28(n=28)和1.62±1.81(n=24)。结果显示口服优福定的5-氟尿嘧啶浓度较复方喃氟啶高,胃癌组口服优福定或复方喃氟啶胶囊后的5-氟尿嘧啶浓度都高于直结肠癌,但都无显著性差别(p>0.05)。

A comparative study of 5-fluorouracil (5-FU) blood concentrations determined by high-perfor-mance liquid chromatography (HPLC) after a single oral dose of UFT or FT207CO containing 200mg of FT207 wascarried out in 138 patients with gastric and colorectal cancer. Among 71 patients receiving UFT, the mean blood concen-trations of 5-FU at 0.5h, 1.0h and 2.0h were 6.69±8.72(n=28), 4.34±3.84(n=32) and 5.76±18.12(n=24) in 38 cases with gastric cancer, and were 4.25±4.34 (n=24), 4.50±5.62(n=27) and 2.39±2.78(n=28)in 33 patients with colorectal cancer, respectively. Among 67 patients receiving FT207CO, the mean blood concentra-tions of 5-FU were 4.89±5.09(n=31), 3.92±3.50(n=33) and 2.59±4.35(n=31) in 36 cases with gastriccancer, and were 2.97±3.55(n=28), 3.32±3.28 (n=28) and 1.62±1.81 (n=24 ) in 31 cases with colorectalcancer, respectively. The data showed that the mean blood levels of 5-FU after oral administration of UFT were slighthigher than those after FT207CO, and the mean blood levels of 5-FU after UFT or FT207CO in gastric cancer groupswere higher than those in colorectal cancer groups, but there were no statistically significant differences (p>0.05).
