脑胶质瘤浸润淋巴细胞体外抗肿瘤活性及产生IL-2水平

摘要 本文动态地观察了9例经 IL-2活化的胶质瘤浸润淋巴细胞(GIL)体外抗肿瘤活性。结果表明 GIL 具有选择杀伤自体胶质瘤细胞特异性,同时也表达 NK 样活性和 LAK 样活性;而对自体淋巴细胞无破坏作用。GIL 产生 IL-2和 TNF 显著高于自体淋巴细胞和正常对照淋巴细胞。研究结果表明 IL—2活化的GIL 可能是较理想的过继性免疫治疗恶性胶质瘤的效应细胞。 In vitro antitumor activity of glioma-infiltrating lymphocytes(GIL)from 9 surgical biopsy specimen of atients with primary brain gliomas were examined ty using 18h LDH release cytotoxicity assay.Results suggested that activity of killing various target cells of GIL increased progressively in cultrue in IL-2,reached the-top in the culture for 3rd week and decreased a little in the culture for 4th week,but there were not defferen(P>0.05).Activity of GIL in the culture in IL-2 of killing autologous glioma cells were significant higher than that of killing allogeneic glioma cells(P<0.05).GIL also expressed the activity of killing K562 cells and Daudi cells and GIL didn' t express the activity of killing autologous lymphocytes.The capacity of GIL secreting IL-2 and TNF was much higher than that of peripheral blood lymphocytes of the patients and control group.Results suggested that GIL might be a new kind of antitumor effector for treatment of patients with brain glioma.
