本文报道了一种高效、方便的脂质体与抗体共价偶联的方法.用过碘酸钠氯化神经节苷指,使之临位羟基变成醛基,与抗体的氨基形成希夫氏碱,然后还原成稳定的共价结构.用 Dextran T40密度梯度离心,把腊质体-抗体复合物与游离抗体分开.碱性磷酸酶(AKP)和辣根过氧化物酶(HRP)与脂质体偶联后,酶活性均保留80%以上.并与戊二醛标记法作了比较.

A efficient and convenient method for covalent binding of antibodies to liposomes was de-scribed.Antibodies were attached to the aldehye groups on the gangliosides by the periodate ox-idation to form schiff—base and the complex was stabilized by reduction.Separate the liposome —IgG complex from unbound IgG using discontinous dextran T40 gradient centrifugation.AKP and HRP were coupled to liposomes,and both enzymatic activity was remained more than 80%.and compared this with glutaraledhyed method.
