补脾肾活血中药对糖尿病大鼠肾皮质糖基化终产物受体mRNA表达的影响 被引量:13

Effects of Bupishenhuoxue Therapy on the mRNA Expression of Advanced Glycation End Products Receptors in the Renal Cortex in the Diabetic Rat

摘要 目的 观察补脾肾活血中药对糖尿病大鼠肾皮质糖基化终产物 (AGEs)含量及其受体mRNA(RAGEmRNA)表达的影响 ,探讨补脾肾活血中药防治糖尿病肾病 (DN)的机制。方法 采用链脲佐菌素 (STZ)左下腹腔内单次注射造成糖尿病大鼠模型 ,随机分糖尿病组、中药组及氨基胍组 ,另设正常对照组。用荧光光谱法测定肾皮质AGEs含量 ,用RT PCR的方法检测肾皮质RAGEmRNA的表达。结果 糖尿病组大鼠肾皮质AGEs含量明显增加 ,RAGEmRNA出现过度表达。中药组治疗 1 2周后肾皮质AGEs含量明显减少 ,RAGEmRNA表达明显下调 ,与糖尿病组比较都有显著差异 (P <0 0 1 )。结论 补脾肾活血中药能够通过减少AGEs在肾皮质的积累 ,下调糖尿病大鼠肾皮质RAGEmRNA的过度表达 。 Objective To observe the effects of Bupishenhuoxue Therapy (BT) on the level of advanced glycation end products (AGEs) and on the mRNA expression of AGEs receptors (RAGE) in the renal cortex in the diabetic rat, and to investigate the mechanism of the effects of BT in preventing and treating diabetic nephropathy (DN). Methods The rat diabetic model was induced by an intraperitoneal injection of streptozotocin in the left lower abdomen of the rat. The model rats were divided into the diabetic group, BT group and aminoguanidine group. And there was a normal control group as well in the experiment. The fluorescence spectrum method was used to detect the level of AGEs, and the RT-PCR technique was used to detect the expression of RAGE mRNA. Results It was found that the level of AGEs was significantly increased and the RAGE mRNA was over expressed, in the renal cortex of the rats in the diabetic group; and the level of AGEs was markedly lowered and the expression of RAGE mRNA was markedly downregulated in the renal cortex of the rats in the BT group after being treated with BT for 12 weeks, as compared with the results from the diabetic group ( P <0 01). Conclusion BT can prevent and treat DN by decreasing the accumulation of AGEs and downregulating the over expression of RAGE mRNA in the renal cortex.
