摘要
目的 :建立Beagle犬血浆中盐酸阿呋唑嗪浓度高效液相色谱测定方法。方法 :采用液 -液萃取法预处理血浆样品 ,分析柱为AglientC1 81 5 0mm× 4.6μm ,5 μm ,流动相为 :乙腈 :0 .0 2 5M磷酸盐缓冲液 ( pH =3) =1 8:82 ,检测波长 :2 4 4nm ,柱温 :2 5℃ ,流速 :1ml /min ,按内标法定量。结果 :盐酸阿呋唑嗪与内标固定物的保留时间为 4.7min和 3.4min ,盐酸阿呋唑嗪血药浓度线性范围为 1 0~ 80 0mg .l- 1 Y =0 .0 2 2 71 5C - 0 .1 1 673(R2 =0 .9999) ,方法回收率为 1 0 2 .2 9% ,日内和日间RSD分别为 1 .0 4~ 4.37%和 2 .0 5~ 6.40 %。结论 :本法操作便捷 ,灵敏度和精密度高 ,重现性好 。
OBJECTIVE To establish an HPLC method for determination of alfuzosin hydrochloride in Beagle dog plasma. METHOD Plasma sample were pretreated by liquid - liquid extaction .alfuzosin hydrochloride and terazosin hydrochloride ( internal stangdard ) were separated on Aglient C18 column (150 mm × 4.6 μm ,5μm ) at 25℃with 0.025mol.l-1 phosphate buffer of 3.0 - actonitrile ( 82 : 18 ) as mobil phase at a flow of 1 ml.L-1,and monitored at 244 nm . RESULT alfuzosin and terazosin was eluted at 3.7 min and 4.8 min after injection , respectively .Calibration curve in Beagle dog plasma was liner Y = 0.022715 C - 0.11673 ( R2 = 0.9999) in the range of 10 to 800ng.ml-1.the analytical recovery was 102.29% ( n = 5 ) ,the within day RSD and between - day were 1.04%~4.37% and 2.05%~6.40% ( n = 5 ),respectively.CONCLUSION The method is simple ,sensitive and reproducible ,and is suitable for the pharcokinetic study of alfuzosin
