摘要
目的 :应用HPLC法测定更昔洛韦中的鸟嘌呤、阿昔洛韦及其他有关物质。方法 :用PhenomenexLunaC18色谱柱 ,以 0 0 2mol/L醋酸铵缓冲液 (pH 4 5 ) 甲醇 ( 95∶5 )为流动相 ,流速为 1ml/min ,检测波长为2 5 4nm。结果 :最低检出限阿昔洛韦为 12 5ng、鸟嘌呤为 2 0ng、其他有关物质以更昔洛韦计为 2 0ng。更昔洛韦与鸟嘌呤、阿昔洛韦及其降解产物的分离度均能达到要求。结论 :该法专属性强 ,结果准确 ,重现性好 ,灵敏度高 ,可用已知杂质法测定更昔洛韦中的鸟嘌呤、阿昔洛韦 。
AIM:A HPLC method was developed for determining the content of acyclovir and guanine and related substances in ganciclovir METHOD:The sample was analysed on C 18 column,with a moble phase of 0 02 mol/L ammonium acetate buffer(pH 4 5) CH 3 OH(95∶5),at the detection wavelength of 254 nm and the flow rate is 1 0 ml/min RESULT:The detection limit of acyclovir was 12 5 ng,guanine was 20 ng and ganciclovir was 20 ng CONCLUSION:This method was simple,accurate,sensitive and good for the determination of acyclovir,guanine and related substances in ganciclovir
