摘要 目的 :探讨白细胞及MMP 9在脑梗死病理生理机制中的作用。方法 :采用细胞免疫组化和RT PCR法 ,检测 2 0例脑梗死患者与 2 0例正常人循环白细胞MMP 9的表达水平。结果 :细胞黄染色为MMP 9表达阳性 ,脑梗死患者循环白细胞均染黄色或澄黄色 ,显示中至强度表达 ,表达部位主要在胞浆 ;与正常人比较 ,脑梗死患者循环白细胞MMP 9mRNA表达明显增强 ,差异有显著意义 ( 2 15± 0 41vs 1 3 2± 0 2 8,P <0 0 0 1)。结论 :提示脑梗死患者循环白细胞处于炎性状态 ,MMP 9表达增强 ,并可能由此导致血浆MMP 9水平增高 ,抑制循环白细胞MMP 9的表达 ,应作为脑梗死防治的重要有效途径之一。 Objective To investigate whether the matrix metalloproteinase-9(MMP-9) expression are increased by peripheral blood Leukocytes in patients with acute cerebral infarction diseases. Methods We measured the MMP-9 expressions in peripheral blood Leukocytes in 20 patients with acute cerebral infarction and in 20 age-and sex-matched healthy subjects with immunocytochemistry and reverse-transcription polymerase chain reaction (RT-PCR). Total RNA was extracted from peripheral blood leucocytes. Amplification of specific sequences was performed by standard PCR methodology, and primers were designed according to sequences obtained from the reference. Results In stroke and clntrol group ,the MMP-9 expressions was 2.15±0.41 and 1.32±0.28 respectively. Compared with the control group, MMP-9 expressions were significantly higher in the stroke group (P<0.001). Conclusion MMP-9 expressions were significantly higher in patients with acute cerebral infarction diseases than in healthy control subjects. Activated leukocytes and leukocytic MMP-9 may have an important role in acute cerebrovascular ischemia and its consequences.
