摘要
本研究利用在苹果(Malus×domesticaBorkh )研究中使用的27个微卫星标记,对云南省农业科学院园艺作物研究所种质资源圃中收集到的部分资源及昆明附近栽培的梨品种进行分析。结果表明这些标记可用于梨品种的研究,但不适于梨的野生近缘植物石屏海棠(MalushupehensisRehd)、腾冲海棠(MalussieboldiiRehd)和子(CotoneastermultifloraBge)的研究。由于本试验电泳时使用的是3%的琼脂糖凝胶,所以得到的等位位点数不如用聚丙稀酰胺凝胶进行电泳的多。根据27对微卫星引物对42个梨品种基因组DNA扩增的结果,可以将供试的42个梨品种分为5群。
Twenty seven microsatellite markers developed from apple,Malus×domestica Borkh.were used for analysis of varieties of pear,collected from resource garden of Horticultural Crop Institute of Yunnan Academy of Agricultural Sciences and gardens around Kunming.The results showed that markers developed from apple can be used for pear,Pyrus pyrifolia analysis,but they did not work well on wild relatives Malus hupehensis Rehd,Malus sieboldii Rehd,and Cotoneaster multiflora Bge.The alleles recorded on an agarose gel were less than that recorded on a polyacrylamide gel.Fourty two varieties were clustered into 5 lineages at the 0.73 by UPGMA method.
