摘要 目的 :应用热休克蛋白72定量分析的方法 ,判定大鼠脑缺血治疗时间窗。方法 :用线栓法制备Wistar大鼠大脑中动脉缺血/再灌注模型。对MRI显示缺血区标本行HSP72免疫组化染色 ,测量阳性表达神经细胞的平均光密度和积分光密度。结果 :随缺血时间的不同 ,永久缺血组平均光密度有显著差异 (F=10.44,P<0.01) ,积分光密度无显著性差异 (χ2=6.99,P>0.05) ;缺血1h再灌注后不同时间平均光密度有显著性差异 (F=33.04 ,P<0.01) ,积分光密度有显著性差异 (χ2=45.09 ,P<0.01) ;缺血2h再灌注后不同时间平均光密度有显著性差异(F=3.00,P<0.05) ,积分光密度无差异 (χ2=5.42,P>0.05)。结论 :大鼠脑缺血的治疗时间窗应在2h以内。考虑到胶质细胞对神经元细胞的保护作用 。 Objective:To determine the therapeutic time window of cerebral ischemia,by quantitative analysis of heat shock protein72(HSP72)following the middle cerebral artery(MCA)occlusion or reperfusion model in the rat.Methods:The male Wistar rats were subjected to MCA occlusion or reperfusion according to the method of intraluminal filament technique.The samples of MRI show ischemic district were stained by HSP72immunohistochemistry,measured the average optical density and integral optical density at positive expression areas.Results:According to time of cerebral ischemia in permanent ischemia group,the average optiˉcal density were significant difference(F=10.44,P<0.01)and the integral optical density no difference(χ 2 =6.99,P>0.05).Reperfusion after cerebral ischemia1hour,the average optical density(F=33.04,P<0.01)and the integral optical density(χ 2 =45.09,P<0.01)were sigˉnificant difference.Reperfusion after cerebral ischemia2hour,the average optical density were significantly different(F=3.00,P<0.05)and the integral optical density no difference(χ 2 =5.42,P>0.05).Conclusion:The therapeutic time window of cerebral ischemia in the rat ought to within2hours.The aegis effect of gliacyte on neurons should be concerned during deterˉmining therapeutic time window that could be prolonged suitably.
