含HSV-TK基因的重组腺病毒的构建及其对肿瘤细胞的抑制作用

摘要 目的 :构建含HSV -TK基因的重组腺病毒AdTK并观察AdTK/GCV系统对体外培养的PG49、H460、MCF -7和HeLa4种肿瘤细胞株的抑制作用。方法 :采用同源重组法构建AdTK ;用MTT法测定细胞生长抑制率。结果 :已成功构建了AdTK ;AdTK/GCV系统对4种肿瘤细胞的生长抑制率随AdTK、GCV浓度及作用时间的增加而升高。结论 Objective:In this research the AdTK was constructed and identified and AdTK/GCV system cytotoxic effect on PG49H460MCF-7and HeLa cell was assayed in vitro.Methods:Using MTT method with AdLacZ being control,cytotoxin effects to PG49,H460,MCF-7and HeLa established tumor cells with AdTK/GCV system were assayed.Results:It is showed that the growth inhibition rate depended on AdTK dose,the concentration of GCV and the time of interaction.Conclusion:The killing effect result revealed that growth inhibition rate increased with the raising of AdTK dose,concentration of GCV and the time of interaction.
