华支睾吸虫病新诊断试剂盒的建立与应用

The Establishment and Use of a New Diagnostic Kit of Clonorchiasis

本所首次试制成功稳定、灵敏、特异、简便和无毒的华支睾吸虫病诊断药盒.本试剂盒具有以下的优越性:(1)采用部分提纯的抗原A作为包被抗原,抗原的蛋白质浓度为0.5μg/ml,其阳性率为97.1％,交叉反应率为4.6％,灵敏性和特异性均优于粗抗原。(2)采用预先包袱抗原,经G液或M液封闭并干燥后于4～10℃条件下至少可保存8周,室温条件下至少可保存两周而仍不失活的平板。(3)采用特殊方法配制能在冰箱中可至少放置半年而不失活的四甲基联苯胺为显色剂。此显色剂不但无致癌作用,而且其显色反应和重复性都优于OPD。 1988年2月药盒开始用于本病的早期诊断,1990年7月此盒用于流行病学调查也得到理想的结果。在343例居民中粪检虫卵阳性者141例,其中ELISA阳性者127例,ELISA阳性率为90.7％。应用本试剂盒于临床诊断和大规模流行病学调查,可早期诊断和治疗患者,因此本盒有很好的经济效益及社会效益。

We have succeeded in preparing a diagnostic kit for ELBA of ctonorchiasis.This kit has some obvious advantages.First of all,by using a partially purified antigen (Antigen A) and TMB,the positive rate of test in the proved patients increased to 97.1%.Secondly,this kit is safe for the use of TMB instead of OPD.The latted has obvious carcinogenic effect.Thirdly,this kit is very convenient for the use of plates coated with antigen A and blocked with solutions M & G before hand.Furthermore,no loss of antigenic activity was observed after storage of the pre-coated and pre-blocked plates in 4℃ for 8 weeks.This kit has been used both in clinical diagnosis and epidemiological survey.In July 1990,343 inhabitants were examined with this diagnostic kit in Ying-Lan Xiang of Jia-Mu-Si city,Heilongjiang province.Stool specimens of those persons were examined 3 times for detecting the ova of clonorchis worms.141 out of 343 persons were ova positive,while 127 cases were ELISA positive.The positive rate for ELISA was 90.07%.
