摘要
①目的 观察穹隆 海马伞切断及去势大鼠脑内不同部位 β淀粉样肽 1 4 0 (Aβ1 40 )表达的变化 ,了解与阿尔茨海默病 (AD)相关的发病机制。②方法 将大鼠分为穹隆 海马伞切断及去势模型组 (模型组 )和假手术组 ,假手术组除不切断海马伞和去除卵巢外 ,其余步骤同模型组。常规处死大鼠取脑组织行Aβ1 40 免疫细胞化学染色 ,观察海马CA1区、皮质区、杏仁复合体区、基底前脑Meynert核等部位Aβ1 40 阳性细胞表达变化。③结果 假手术组大脑各观察区有少量的Aβ1 40 表达。与假手术组比较 ,模型组大脑海马CA1区、大脑皮质区、杏仁复合体区Aβ1 40 阳性细胞数明显增多 (t=3.96 9~ 6 .6 77,P <0 .0 1)。④结论 穹隆 海马伞切断加去势可致大鼠脑内多部位Aβ1 40 表达增多 ,此可能与老年性痴呆病理改变有关。
ObjectiveTo observe the expressive varieties of β-amyloid 1-40(Aβ 1-40) in different sections of the rat brain with fimbria/fornix transection and ovaries and to study the pathogenic mechanisms related to Alzheimer's disease(AD). Me- thodsHealthy female Wistar rats were randomly divided into the model group and control group with five rats in each group. The fimbria/fornix transection and ovaries cutting were simultaneously performed for the animal model. The rats were executed and the brain taken conventionally. The Aβ 1-40 immunocytes were stained by using an immunohistochemical technique. The expressions of the Aβ 1-40 masculine cells in the hippocammpi CA1, cortex, amygdala, Meynert of the basal forebrain were observed. Results A little Aβ 1-40 was expressed in the observed sections of the brain in the control group, while the Aβ 1-40 masculine cells in the model group were significantly increased than those in the control group in the hippocammpi CA1, cortex, and amygdala (t=3.969- 6.677,P<0.01). ConclusionFimbria/fornix transection and ovaries cutting can cause the increase of Aβ 1-40 expression in many sections of the brain, which might be related to the cognitive disturbance due to Alzheimer's disease.
