生物素-亲和素酶联免疫吸附试验检测弓形虫抗体

摘要 本文用生物素标记葡萄球菌A蛋白,用辣根过氧化物酶标记亲和素,建立了生物素-亲和素酶联免疫吸附试验,用于测定猪血清中抗弓形虫抗体。本方法比常规PPA-ELISA敏感约5倍,比间接血凝法敏感约25倍,特异性和稳定性较好,可用于检测多种动物抗弓形虫抗体,是一种理想的方法。 The biotin-avidin system (BAS) and staphylococcal protein A (SPA) were incoporated into immunoenzymatic techniques,with biotin labelling SPA and horseradish (HRP) labelling avidin to develop a very sensitive and relatively simple and specific assay for detection of Toxoplasma-ticantibodies,biotin-avidin enzymelinked immunosorbent assay(BA-ELISA).The results indica-ted that BA-ELISA was about 5 times more sensitive than PPA-ELISA and 25 times than IHA in the detection of serum antibodies of Toxoplasma The co-efficients of variation (C.V.) were ranged from 5 to 15%.The reaction was specifically and evidently inhibited by antigens.This method could be utilized to detect serum antibodies from animals.
